I agree with @Applebaby12 on this poll, his "partnership" with Cthulhu definitely was the most destructive.

There aren't any I can think of off the top of my head.

Chef's death was the saddest for me. I think it really marked the end of "Classic" South Park and it's really a bummer that he had to die the way he did.
