I know most of what I'm about to write doesn't quite pertain to the question, but given what I consider to be common misinterpretations of Butters (including those like in the review you mentioned) I'm just going to talk about him for a bit. It Will be long, stupid, MY OWN OVERLY DETAILED INTERPRETATION, and contains multiple links for Tumblr. Not that anyone cares.
FOR THE RECORD, I do think Butters has actually changed a lot over the course of the series. But not in the way some think.

Now, for the misinterpretations. For one, Butters has always had a mean streak just like all the other kids, long before season 20 or even season 15 as some argue. Bullying Mark Cotswolds, picking on Kyle and later Cartman (pretty justified with the latter but I digress) once they're respectively deemed highly unattractive, the specifics of how he acted in The Breast Cancer Show ever. One of his favorites to act mean towards is Kyle.
Another thing is that Butters has been sexist pretty much the entire series, starting (to my memory) with him being... spirited about beating the girls in "Cartman's Silly Hate Crime 2000". He wanted to "show those skanky hoes who's who". This isn't an OOC moment either as there are many more examples of him being sexist over the series. My personal theory is that it's rooted in his neurotic, abusive mother, among a few other factors,
Finally, to add on to your point that with all he has to deal with lashing out makes perfect sense. Let's not forget that's far from the first time he's lashed out. Going Native or Professor Chaos, anyone?
As for my actual opinion, I'm pretty neutral. It's not the most entertaining thing ever, but I'm not disgusted by the plot or anything.

Under the context that they simply “meet”, they probably wouldn’t do anything. Judging by general in-universe thoughts on the characters, Shelly likely thinks Nelly is a turd and Nelly thinks Shelly is just Stan’s bitchy sister.

Here are my opinions I guess.
While I certainly understand you on Butters, I personally feel he’s one of the most developed, 3D characters in the show and am very much looking forward to his screentime in the future.
I just think Kyle is neat!
He admittedly wasn’t the best-written character (in fact I wouldn’t say it’s much of a stretch to say he was meant to be a bit too perfect personality-wise) but I love Pip. I think he was a cute and endearingly simple if one-note foil of the early seasons.
I may have a pretty mixed relationship with her character (not helped by Wendy simps who act like she’s a damn Mary Sue), but I feel many people hate on Wendy for kind of dumb reasons. For one, yeah Stan isn’t the best boyfriend, I don’t really blame her and in general after the early days I find Stendy to be mutual shit in canon. Also, come on. We all know the making fun of breast cancer was just the last straw from Cartman and that he more than had that coming. In fact, I (and many others according to TVtropes) feel like that episode kind of redeems her as a character. Also, why is it that some people discredit Wendy for that yet cheer for Kyle beating Cartman’s ass constantly? Also, let’s not act like our main characters haven’t all either murdered or tried to murder in some form while in character. I love the main 4/5, but honestly she’s a fairly good kid at least compared to those assholes.
Carol is a pretty OK character. She’s a really horrible mother and clearly not the best person, but she never really bothered me.
I’m with @Sega&Nicktoons fan 3000 on Liane, she is one of the worst parents and a pretty bad person in my eyes (does everyone forget she forced Cartman to watch her fuck someone at one point?) and I personally find her somewhat irritating.
All in all, I think I’m neutral in regards to Garrison. He’s had funny times, he’s had shitty times.
We shit on Mr. Hankey because he’s another piece of shit, easy.
In all honesty, Cartman is one of the reasons I think South Park’s time is likely over. All in all I really like Cartman, but he hasn’t been so great since Season 20 imo. His whole purpose is being this horrible, horrible child who is the last person you should really side with, and he’s perfect that way. (Hence why I very much disliked his characterization in Season 20 and the latest special.) And if Cartman being horrible is now so bland that people actually want him to change, I think his time is about up.

Cartman’s early seaons character was basically being an ignorant brat who gets fucked with, is fat, and has a number of catchphrases. Because of that, I imagine some sort of running gag would’ve been spawned/enabled regarding the relationship with his sister (likely involving something the sister wouldn’t like too much), and that would likely be said sister’s main purpose.
